Microglia are central nervous system innate immune cells. Although numerous methods for isolating microglia from the brain have been developed, the method of dissociation and cell separationcan have a significant impact on the function of these highly dynamic cells.

We present an optimized Pluribead protocol for isolating CD11b+ cells (microglia) from mouse or human brain tissue. Isolated microglia can be used to model diseases with neuroinflammatory components in order to find potential therapeutics.

Tissue dissociation into single-cell suspensions with high cell viability and low cell debris is required for consistent cell separation, cell culture, and flow cytometry analysis.

PluriBead Technology
PluriBead separates cell mixtures using non-magnetic monodispersed microparticles (beads). Their surface is coated with monoclonal antibodies (mAb) that are directed at specific structures on the target cell's surface. During incubation, the target cells in suspension will bind to the pluriBeads and can then be separated from the suspension using a pluriStrainer (size exclusion).

Because the beads are larger than the cells, they cannot be phagocytized. There is no need for pretreatment of the blood (e.g. density centrifugation, erythrolysis, or other target concentration).
