Twinspin: Pre-Filled Centrifuge Tube To Support Density Gradient Centrifugation

Posted July 5, 2022 by Uberstrainer

Are you searching for centrifugation tubes that offer incredibly fast, simple workflow? Let’s explore the top Pre-filled centrifuge tube, TwinSpin in detail.

For optimal separation of peripheral blood mononuclear cells from whole blood and bone marrow, TwinSpin centrifugation tubes pre-filled with Density Gradient Medium (DGM) can be used. The TwinSpin is made up of a standard 15 ml bottle and an inner tube. The inner tube is submerged in the DGM and has an open bottom.

Pipette diluted anticoagulated blood or bone marrow into the TwinSpin. Inside the inner tube, the sample is placed on top of the DGM. Depending on the DGM, leukocytes, lymphocytes, and PBMCs are separated from unwanted erythrocytes and granulocytes during Density Gradient Centrifugation. As a result, the interphase is enriched in target cells above the DGM.

The erythrocytes will settle out of the inner tube at the bottom of the outer tube via the density gradient media. Simply remove the inner tube once the separation is complete. The elastic cap serves as a valve. By removing the cap, the collection tube transforms into a pipette. Drop by drop, the contents can be collected.

Prefilled with DGM of your choice

1. Leuko Spin - Used to separate leukocytes from whole blood or bone marrow.
2. PBMC Spin - Used to separate PBMC from whole blood or bone marrow.
3. PBMC 24+ Spin - For the isolation of PBMC from whole blood or bone marrow older than 12 hours.
4. PLT Spin - Used to separate platelets from whole blood or bone marrow.

Workflow

1. Remove the stopper. Sample pipette on top DGM
2. Close tightly with an elastic cap (provided separately)
3. Spin for 15 minutes at 800xg at room temperature with the brake on.
4. Unscrew and remove the dropper
5. Hold down the cap until DGM exits.
6. Place target cells in a separate tube.

Uses of TwinSpin
• Best Price-to-Value Ratio
• Minimal handling equals minimal cell stress.
• No more shaky pipetting.
• Ideal for low cell counts.
• Customized pre-filling is available.

Facts & Features of TwinSpin
• Combines density gradient cell separation with pipetting precision to enrich cells with maximum yield and viability.
• Compatible with standard density gradient centrifugation protocols
• Enrichment of untouched specific cells in combination with pluriSpin negative cell separation from whole blood, buffy coat, or cord blood
• Useful for preparing samples for magnetic cell separation.
• There is no need for any special training or equipment.

Select your separation method based on the distinct differences between your target particle and the undesirable impurities.
